Combination of 2,4-Dichlorophenoxy Acetic Acid and Benzyl Adenine Concentration on The Growth of Aglaonema sp. with Tissue Culture Technique
Abstract
Plant seeds can be produced in large quantities quickly, with quality, uniform seed size, and free from disease, one of which is by tissue culture. The composition of the media determines the success of tissue culture, including the concentration of plant growth regulators. Exogenous plant growth regulators of the auxin and cytokinine groups that are widely used in tissue culture media are 2,4-Dichlorophenoxy Acetic Acid (2,4-D) and Benzyl Adenine (BA). This study aims to determine the effect of giving a combination of 2,4-D and BA concentrations on the growth of Aglaonema sp. with tissue culture. The study was conducted at the Biotechnology and Tissue Culture Laboratory, Faculty of Agriculture, Universitas Muhammadiyah Jakarta. The study used a Completely Randomized Design (CRD) Factorial, with 2 factors, namely, factor 1: 0.1 mg/l 2,4-D, 0.3 mg/l 2,4-D, and 0.5 mg/l 2,4-D; factor 2: 8 BA, 9 mg/l BA, and 10 mg/l BA. The results showed that the administration of a combination of 2,4-D and BA concentrations had no effect on the growth of Aglaonema sp., but the administration of a single concentration of BA had an effect on the percentage of living explants. The administration of a single dose of 8 mg/l BA had a very significant effect on the percentage of living explants, while the administration of a combination of 0.1 mg/l 2,4-D + 8 mg/l BA concentrations gave the best results and was significantly different from the percentage of living explants.
