PENGARUH PENGGUNAAN VITAMIN E DALAM PENGENCER SUSU SKIM PADA SPERMA BEKU TERHADAP PERSENTASE MOTILITAS DAN PERSENTASE SPERMATOZOA HIDUP DOMBA PRIANGAN

ABSTRACT
The objective of this research is to know the optimal concentration level of vitamine E in the skimmed milk dilution against Priangan sheep’s motility and lived spermatozoa presentation. The research was done in the Laboratory of Artificial Insemination Institution, Lembang, Bandung, from November 2017 up to January 2018. In the experiment, spermatozoa from five of Priangan sheep collected by artificial vagina and diluted with skimmed milk, each dilution added by different dose of 10, 20, 30, 40 μg vitamine E and without vitamine E per mililiter for control. The experimental method used was Completely Randomized Design continued by Duncan multiple rank test 5% significant. Observed variable was motility and lived spermatozoa presentation measured after dilution and equilibration, and after thawing with include percentage of motility, lived. The result of experiment showed that percentage of motility and lived spermatozoa after dilution were not significantly after treatment (P>0.05), after equilibration and thawing resulted the highest percentage of motility and lived in the treatment of E30 (72.00 and 54.00 %), and (82.20 and 67.60%) significantly different compared to the other treatment (P<0.05).
ABSTRAK
Penelitian ini bertujuan untuk mengetahui tingkat konsentrasi vitamin E yang optimal pada pengencer susu skim terhadap persentase motilitas dan persentase spermatozoa hidup pada sperma beku Domba Priangan. Penelitian ini dilakukan di laboratorium Balai Inseminasi Buatan, Lembang, Bandung dari bulan November 2017 sampai bulan Januari 2018. Pada percobaan, spermatozoa dari lima ekor Domba Priangan ditampung dengan vagina buatan dan diencerkan dengan susu skim, masing-masing pengencer diberi vitamin E dengan dosis yang berbeda, yaitu tanpa vitamin E sebagai kontrol, 10, 20, 30 dan 40 ïg vitamin E per mililiter. Metode eksperimental menggunakan rancangan dasar Rancangan Acak Lengkap, yang dilanjutkan uji jarak berganda Duncan taraf nyata 5%. Peubah yang diamati adalah kualitas spermatozoa yang diukur setelah pengenceran, setelah ekuilibrasi, serta setelah pencairan kembali yang meliputi persentase motilitas, persentase hidup spermatozoa. Hasil percobaan menunjukkan bahwa persentase motilitas dan persentase spermatozoa hidup pasca pengenceran tidak berbeda nyata antar perlakuan (P>0,05). Pada pasca ekuilibrasi dan pasca pencairan kembali yang menghasilkan persentase motilitas dan persentase hidup tertinggi pada perlakuan E30 (72,00 dan 54,00 %) dan (82,20 dan 67,60 %) berbeda nyata dibandingkan dengan perlakuan yang lain (P<0,05).
